报告题目:Combining multiple SNPs in Mendelian randomization studies with continuous outcomes

摘  要:Mendelian Randomization (MR) uses genetic variants as instrumental variables (IVs) to estimate causal effects of exposure on outcomes. The core conditions of instrumental methods require instrumental variables 1) are associated with exposure; 2) have no direct pathway to outcomes; 3) are not associated with any confounder that affects both exposure and outcome. In practice due to pleiotropy effect or other genetic factors such as linkage disequilibrium and population stratification, the core conditions 2) and 3) may be violated and IVs become invalid.  In this talk, we compare several recently developed methods, including MR-Egger, MR-median, sisVIVE used in MR when not all IVs are valid. We discuss the advantage and drawbacks of each method and illustrated by simulation &UKHLS data investigating causal effect of BMI on several social outcomes.


报告人简介:

Dr. Bao Yanchun  got her BSc in statistics at Yunnan University and her MSc in Statistics (with Professor Shuyuan He) at Peking University. She studied for her PhD at University of Manchester (with Professor Janxin Pan) and now works at ISER, University of Essex. She is interested in Bioinformatics research areas and statistical methods for social-health research, including Hidden Markov Model (HMM) for ChIP-sequence and Mythelation data, Instrumental variables methods for Mendelian Randomization, survival analysis and longitudinal analysis.
